I would have to say its when Bond kicks the car of the cliff. A pretty cold kill especially for Roger:
I'd say any moment when Bond actually kills in cold blood. Anything which echoes the character as Fleming intended, rather than the increasingly cartoony comedic iterations.

Also this particular moment:

GW549H234

Forget your .50cal Desert Eagles and whatever other manly movie guns. This is the Beretta 950 Jetfire, one of the tinest and unmanly pistols around... yet Bond looks like he's gonna really go **** someone up with it!!

There's a bit in a recent one - I think Spectre - it's just after he's had his brain drilled when they are escaping the compound and he just goes total efficient killing machine. A lot of people really don't like it because they see it as unrealistic that he's so good, and it also goes against the 'vulnerable Bond' we saw in Casino Royale.
But I see it as a deliberate juxtaposition - this is what he's become through his development as a 00 since the reboot, this is how dangerous he is now - and I think it works really well.

The "Bond. James Bond." at the end of Casino Royale is done brilliantly too. Many of the best older film moments are already cited above. Union Jack parachute for instance.

The Spy Who Loved Me, skiing over the cliff, free falling for a bit, Union Jack parachute, Bond theme music. Marvellous!
